IGF-1 Released During Osteoclastic Bone Resorption Induces Osteoblast Differentiation from Bone Marrow Stromal Cells

چکیده

Homeostasis of skeletal tissues is maintained by a well-balanced regulation of bone formation and bone resorption (1;2). This process is called remodeling, in which osteoblasts are involved in the bone formation process and osteoclasts are essential for bone resorption. During the remodeling process, bone resorption by osteoclasts acts as a trigger to stimulate bone formation by osteoblasts, a step called coupling (3). However, the factor(s) that mediate bone formation signals from osteoclasts to osteoblasts are not wellunderstood. Several factors, such as TGF-β, have been reported as possible candidates (4), but no direct evidence exists thus far.

TGF-β Inhibition Restores Terminal Osteoblast Differentiation to Suppress Myeloma Growth

BACKGROUND Multiple myeloma (MM) expands almost exclusively in the bone marrow and generates devastating bone lesions, in which bone formation is impaired and osteoclastic bone resorption is enhanced. TGF-beta, a potent inhibitor of terminal osteoblast (OB) differentiation, is abundantly deposited in the bone matrix, and released and activated by the enhanced bone resorption in MM. The present ...

Thiazolidinediones Inhibit Osteoclast-Like Cell Formation and Bone Resorption in Vitro1.

Osteoblasts and adipocytes are derived from common bone marrow stromal cells that play crucial roles in the generation of osteoclasts. Activation of peroxisome proliferator-activated receptor-gamma (PPARgamma) induces adipogenic differentiation of stromal cells; however, whether this would affect osteoblast/osteoclast differentiation is unknown. Thus, we examined the effects of the thiazolidine...

Ghrelin is an Osteoblast Mitogen and Increases Osteoclastic Bone Resorption In Vitro

Ghrelin is released in response to fasting, such that circulating levels are highest immediately prior to meals. Bone turnover is acutely responsive to the fed state, with increased bone resorption during fasting and suppression during feeding. The current study investigated the hypothesis that ghrelin regulates the activity of bone cells. Ghrelin increased the bone-resorbing activity of rat os...

Induction of Mineralized Nodule Formation in Rat Bone Marrow Stromal Cell Cultures by Silk Fibroin

Background: Silk fibroin is a suitable protein for osteogenesis by inducing markers of bone formation in the cultures of osteoblasts, so we examined the ability of this protein to induce mineralized nodules in the rat bone marrow stromal cell cultures. Methods: Bone marrow stromal cells obtained from 4 to 6 weeks old Spruge-Dawely male rats were grown in primary culture for seven days and then ...

Low-frequency vibration treatment of bone marrow stromal cells induces bone repair in vivo

Objective(s):To study the effect of low-frequency vibration on bone marrow stromal cell differentiation and potential bone repair in vivo. Materials and Methods:Forty New Zealand rabbits were randomly divided into five groups with eight rabbits in each group. For each group, bone defects were generated in the left humerus of four rabbits, and in the right humerus of the other four rabbits. To t...
